Identification of rigid counterparts for common flexible scaffolds is crucial to the advancement medicinal chemistry. Herein, synthesis new bicyclo[2.1.1]hexanes act as rigidified cyclopentanes described.

A synthetic approach to highly functionalized cyclopentanes that incorporate adjacent secondary and tertiary alcohols is described. These systems, prepared in one step by a ketyl radical cyclization of β-disubstituted acrylates, represent the core of the marine prostanoid family of natural products.

A highly enantioand diastereoselective intramolecular C–H insertion reaction of α-diazo esters has been achieved with the use of dirhodium(II) tetrakis[N-phthaloyl-(S)-tert-leucinate] as a catalyst, providing exclusively cis-2-arylcyclopentane-1-carboxylates in up to 95% ee with no evidence of alkene formation.
